Trample is a creature ability that allows the creature to "trample" over the block creatures and still do damage to the target opponent. Trample is generally saved for big creatures with high power ratings.

Example: Ball Lightning is a 6/1 with trample. If a 1/1 blocks Ball Lightning, then the player will still take 5 damage as the creature's toughness was 1, taking 1 damage off the damage being dealt.
